Ofloxacin belongs to a class of drugs called quinolone antibiotics. It works by stopping the growth of bacteria. This antibiotic treats only bacterial infections.
Ofloxacin containing ofloxacin is a synthetic 4-fluoroquinolone antibacterial agent with bactericidal activity against a wide range of Gram-negative and Gram-positive organisms. Ofloxacin is thought to exert bactericidal effect by inhibiting DNA gyrase, an essential enzyme that is a critical catalyst in the duplication, transcription and repair of bacterial DNA. Following oral administration ofloxacin is rapidly and well absorbed from G.I. tract. The bioavailability of ofloxacin in the tablet formulation is almost 100%. It is widely distributed into body tissues and fluid. The half-life is about 4.6 to 6.9 hours. Up to 80% of an oral dose of ofloxacin is eliminated in the urine as the parent compound within 48 hour.
OPHTHALMIC Bacterial conjunctivitis: Adult: As 0.3% ophth solution: 1-2 drops in the affected eye instilled every 2-4 hr for the 1st 2 days then 4 times daily for the next 5 days of treatment. Child: >1 yr: 1-2 drops in the affected eye instilled every 2-4 hr for the 1st 2 days then 4 times daily for the next 5 days of treatment. Bacterial corneal ulcer: Adult: As 0.3% ophth solution: Days 1-2: Instill 1-2 drops into the affected eye every 30 min while awake. Awaken approx 4 & 6 hr after retiring & instill 1-2 drops. Days 3-7 (or 9 if necessary): Instill 1-2 drops hrly while awake. Thereafter: Instill 1-2 drops 4 times daily until condition fully resolved. Child: >1 yr: Days 1-2: Instill 1-2 drops into the affected eye every 30 min while awake. Awaken approx 4 & 6hr after retiring & instill 1-2 drops. Days 3-7 (or 9 if necessary): Instill 1-2 drops hrly while awake. Thereafter: Instill 1-2 drops 4 times daily until condition fully resolved.
Antacids containing magnesium, aluminium or calcium may decrease absorption of ofloxacin. Iron or Zinc may decrease oral absorption of ofloxacin.
Contraindicated for the treatment of certain sexually transmitted diseases by some experts due to bacterial resistance.
Headache. Dizziness. Upset stomach or throwing up. Not able to sleep.
This drug should be used during pregnancy only if the benefit outweighs the risk to the fetus; according to some authorities, use is contraindicated. Use is considered acceptable with monitoring of the infant for possible effects on the gastrointestinal flora (e.g., diarrhea or candidiasis [thrush, diaper rash]); avoiding breastfeeding between 4 to 6 hours after maternal dosing should decrease the infant's exposure to this drug in breast milk. -According to some authorities, use is contraindicated. -According to other authorities, a decision should be made to discontinue breastfeeding or discontinue the drug, taking into account the importance of the drug to the mother.
