Indicated for the relief of acute and chronic pain, including post-operative pain, dental pain, musculo-skeletal pain, acute traumatic pain and cancer pain.
Nefopam Hydrochloride is a centrally acting analgesic with a rapid onset of action. The main site of action appears to be in the central nervous system both at the brain and spinal levels. In vitro experiments have shown Nefopam to inhibit the re-uptake of various catecholamines (including noradrenaline, serotonin and dopamine). It is possible that the mechanism of action of nefopam is at least in part by altering the levels of these neuromodulators in the brain and at the spinal level.
INTRA VENOUS Moderate acute & chronic pain: Adult: 20 mg by slow IV inj every 4 hr. Max 120 mg daily. Patient should be lying down while receiving inj & should remain so for 15-20 min.
History of convulsive disorders. Glaucoma, urinary retention, hepatic or renal impairment. Elderly. Pregnancy.
Nausea, vomiting, sweating, drowsiness, insomnia, urinary retention, dizziness, hypotension, tremor, paraesthesia, palpitations, lightheadedness, nervousness, mental confusion, blurred vision, headache, dry mouth, angioedema, tachycardia, euphoria, hallucinations, convulsions, temporary pink discolouration of the urine.
Nefopam should NOT be taken during pregnancy or while breast-feeding. If you are pregnant or breast-feeding, think you may be pregnant, or are planning to have a baby, ask your doctor or pharmacist for advice before taking this medicine.
Nefopam toxicity is manifested by neurological symptoms (convulsions, hallucinations, agitation) and cardiovascular response (tachycardia with hyperdynamic circulation). Supportive treatment is suggested including gastric lavage, forced emesis and diuresis. Oral administration of activated charcoal may help prevent absorption. Convulsions and hallucinations may be controlled with diazepam. Beta-adrenergic blockers may be of use in controlling the cardiovascular complications.
