Treatment of uncomplicated acute illness due to influenza infection in patients ?2 weeks of age who have been symptomatic for ?2 days. Prophylaxis of influenza in patients ?1yr of age.
Neuraminidase Inhibitor; Antiviral
Oseltamivir is an ethyl ester prodrug requiring ester hydrolysis for conversion to the active form, Oseltamivir carboxylate. The proposed mechanism of action of Oseltamivir is via inhibition of influenza virus neuraminidase with the possibility of alteration of virus particle aggregation and release.
ORAL Influenza A&B: Adult: 75 mg bid for 5 days. Initiate treatment w/in 2 days of onset of symptoms. Child: 1-12 yr: Wt 15kg: 30mg bid; >15-23 kg: 45mg bid; >23-40 kg: 60 mg bid; >40 kg: 75 mg bid. Initiate treatment w/in 2 days of onset of symptoms. Duration of treatment: 5 days. Prophytaxis of flu A&B: Adult: 75 mg once daily for 10 days. Initiate treatment w/in 2 days of exposure. For community outbreak: May continue dosing for up to 6 wk. Child: 1-12 yr: Wt 15 kg: 30 mg once daily; >15-23 kg: 45 mg once daily; >23- 40 kg: 60 mg once daily; >40 kg: 75 mg once daily. Initiate treatment w/in 2 days of contact w/ an infected individual. Duration of treatment: 10 days.
Information derived from pharmacology and pharmacokinetic studies of Oseltamivir suggests that clinically significant drug interactions are unlikely. Co-administration with amoxicillin does not alter plasma levels of either compound.
Hypersensitivity. Severe renal impairment. Moderate renal impairment. Pregnancy & lactation. Chronic cardiac disease, hepatic impairment. resp disease, immunocompromised patients.
Nausea, vomiting, abdominal pain, bronchitis, insomnia, vertigo, diarrhoea, dizziness, headache, cough, fatigue, unstable angina, anaemia, pseudomembranous colitis, pneumonia, pyrexia, peritonsillar abscess, elevated liver enzymes, hepatitis, aches & pains, dyspepsia, rhinorrhoea, upper respiratorytract infections. In children: Vomiting, other GI disturbances, asthma, bronchitis, conjunctivitis, dermatitis. epistaxis, ear disorders, otitis media, lymphadenopathy, pneumonia, sinusitis. Anaphylaxis, severe dermatologic reactions.
This drug should be used during pregnancy only if the benefit outweighs the risk to the fetus. Caution is recommended; benefit should outweigh risk during breastfeeding.
At present there has been no experience of over dose. Single dose of up to 1000 mg of Oseltamivir have been associated with nausea and/or vomiting.
Store at 25°C. Reconstituted susp: Store between 2-8°C for up to 17 days or at 25°C for up to 10 days. Do not freeze.
