Nikethamide is a stimulant which mainly affects the respiratory cycle. It was used in the mid-twentieth century as a medical countermeasure against tranquilizer overdoses, before the advent of endotracheal intubation and positive-pressure lung expansion.
Nikethamide also known as nicotinic acid diethylamide is a central & respiratory stimulant. It has direct stimulatory action on CNS, increases the frequency & depth of breathing. It has no immediate impact on the cardiovascular system. It is extensively metabolized in liver. All metabolites & a small amount of unchanged drug is excreted in urine.
Anesthetic, sympathomimetics, MAOIs, neuromuscular blockers, bupropion.
Porphyria, epilepsy or other convulsive disorders, cerebral oedema, cerebrovascular accident, head injury, acute severe asthma, physical obstruction of the airway, severe hypertension, ischaemic heart disease, hyperthyroidistn, phaeochromocytoma. Less severe degrees of HTN or impaired cardiac reserve; liver dysfunction.
Dyspnoea, resp problems; muscular, GI, CNS & CV effects. Convulsions.
Use in pregnancy & lactation is safe.
