Indicated for the suppressive treatment and treatment of acute attacks of malaria due to Plasmodium vivax, P. malariae, P. ovale, and susceptible strains of P. falciparum. It is also indicated for the treatment of discoid and systemic lupus erythematosus, and rheumatoid arthritis.Special use in COVID-19
ORAL Rheumatoid arthritis; Systemic lupus erythematosus: Adult: Initially, 400 mg daily in divided doses. Maintenance: 200- 400 mg/day. Max: 6.5 mg/kg/day or 400 mg/day whichever is lower. Child: Up to 6.5 mg/kg/ day or 400 mg/day whichever is lower. Lowest effective dose should be used. Prophylaxis of malaria: Adult: 400 mg every 7 days. Begin 2 wk before exposure, continue for 4-6 wk after leaving the endemic area. Child: 6.5 mg/kg once wkly. Max: 400 mg/dose. Acute malaria: Adult: Initially, 800 mg followed by 400 mg 6-8 hr later, then a further 400 mg on each of the succeeding 2 days. Child: 13 mg/kg; followed by 6.5 mg/kg 6 hr later & repeat dose on the 2nd & 3rd days.

Day 2-10 : 200mg – TID
For Childern> 9yrs
Day 1: 11mg/Kg – BID
Day 2-10: 5.5mg/KG – TID

400 mg BD on day 1
Followed by
400 mg once weekly for next seven weeks (To be taken with food)
Retinal or visual field changes, known hypersensitivity. Long-term use in children. Impaired liver or renal function, severe GI disorders, porphyria, psoriasis, neurological disorders esp a history of epilepsy, myasthenia gravis, G6PD deficiency, pregnancy, lactation. Monitor CBC in patients receiving prolonged therapy. Perform baseline & periodic 6-mth eye exams, test periodically for muscle weakness.
Retinopathy, hair loss, photosensitivity, tinnitus, myopathy (long-term therapy). Psychosis, seizures, leucopenia & rarely aplastic anaemia, hepatitis, GI upsets, dizziness, hypokalaemia, headache, pruritus, urticaria, difficulty in visual accommodation. Loss of hair, bleaching of hair pigment, bluish-black pigmentation of the mucous membranes & skin, photosensitivity, tinnitus, reduced hearing, nerve deafness, neuromyopathy, & myopathy, including cardiomyopathy.
Use should be avoided during pregnancy except in the suppression or treatment of malaria the judgment of the physician the benefit outweighs the risk. Some experts do not recommend breastfeeding during therapy with this drug. Infants exposed to this drug during breastfeeding receive only small amounts of the drug. In a small number of infants up to at least 1 year of age, careful follow-up found no adverse effects on growth, vision, or hearing.
