Treatment of irritable bowel syndrome with diarrhea (IBS-D).
Mu-opioid receptor agonist.
Eluxadoline is a mu-opioid receptor agonist, kappa opioid receptor agonist & delta opioid receptor antagonist. Eluxadoline works directly in intestine to reduce peristalsis, also makes the nerves in intestine less sensitive to stimulation.
Adult: Take with food. 100mg twice daily. Patients with mild or moderate hepatic impairment, receiving concomitant OATP1B1 inhibitors (see Interactions), or unable to tolerate 100mg dose: 75mg twice daily. Children: Not established.
Oral Contraceptive, Cyclosporine, Probenecid, Rosuvastatin.
Without a gallbladder. Biliary duct obstruction. Sphincter of Oddi disease or dysfunction. Alcoholism, alcohol abuse/addiction, or >3 alcoholic drinks/day. History of pancreatitis or structural disease of the pancreas (including pancreatic duct obstruction). Severe hepatic impairment. History of chronic or severe constipation or sequelae from constipation. Mechanical GI obstruction.
Constipation, nausea, abdominal pain; sphincter of Oddi spasm, pancreatitis, hypersensitivity reactions (discontinue if occur).
There are no studies with Eluxadoline in pregnant women that inform any drug-associated risks. The background risk of major birth defects and miscarriage for the indicated population is unknown.
Store in a cool & dry place, protect from light. Keep out of the reach of children.
