Cinnarizine is predominantly used to treat nausea and vomiting associated with motion sickness, vertigo, Ménière's disease, or Cogan's syndrome. In fact, it is one of only a select few drugs that has shown a beneficial effect in the chronic treatment of the vertigo and tinnitus, associated with Meniere's disease.
Calcium-channel blocker; Antiemetic
Cinnarizine acts as an antihistamine, labyrinthine sedative and a peripheral antivasoconstrictor. Cinnarizine is a selective calcium antagonist, inhibiting the influx of Ca2+ intracellularly. It prevents the Ca2+ dependent contraction of arterial smooth muscle by inhibiting Ca2+ influx through smooth muscle calcium channels and thereby, improves vestibular symptoms and prevents peripheral arterial disease
ORAL Motion sickness: Adult: 30 mg taken 2 hr before travel & 15 mg every 8 hr during the journey if necessary. Child: 5-12 yr: 1/2 of adult dose. Vertigo & vestibular disorders: Adult: 30 mg tid. Child: 5-12 yr:1/2of adult dose.
No drug interactions have been seen with Cinnarizine when administered concomitantly with antihypertensives, diuretics, anticoagulants or hypoglycaemics.
Proven hypersensitivity to the drug. Parkinson’s disease. Childn & neonates. Hypotension; pregnancy; lactation; elderly. May impair ability to drive or operate machineries.
Extrapyramidal symptoms sometimes associated w/ severe depression. Drowsiness, headache, GI upsets, unsteadiness, headache; rarely skin & hypersensitivity reactions, dry mouth, blurred vision, urinary difficulty or retention, constipation & increased gastric reflux, fatigue. Hypollpidaemic effect.
During pregnancy and lactation (breastfeeding) Cinnarizine Quality Pharmaceutical Laboratory usage is possible only in exceptional cases, when the expected benefit to the mother far outweighs the potential risk to the fetus or infant.
